Mostly it is open Wednesday till Sunday, 11 AM till 5 PM.

To get there by public transport will take about 3 or 4 hours, depending on your route, and it will cost about 40 Euro for a same day return ticket, but it is a difficult journey to explain, best look for yourself on http://www.9292ov.nl/

In addition to the answer above: do expect to have to travel for quite some time to get there. You will have to take a train to Schiedam where you have to get on the metro to Spijkenisse. There you need to transfer to a bus to Hellevoetsluis where you have to transfer on another bus again and my guess is that you have to transfer to another bus later on as well.
You can also take the train south to Vlissingen but this will take a long time as well, plus you will need to travel back up north again from Vlissingen so either way you will end up taking a bus.
It's not easy to get around in the province of Zeeland by public transportation because there are no trains going through this province.
Plan you trip to the Delta Expo very well and I recommend you to rent a bungalow or room somewhere in Zeeland so you won't have to do all the travelling in one day.
